While the compound microscope would be more effective for viewing the frequency of the stoma, a stereo dissecting microscope can also be used for this purpose. Procedure: Remove a leaf from the plant and place it flat on the table. Students may need to repeat this step several times to get a thin enough slice. Typically, the stomata are bean shaped and will appear denser darker under the microscope. Have professional leaf cross-section slides available for them to use if their procedures fail.
I think the first step is to make careful hand sections. With practice you can get excellent sections of living material.
Put a piece of leaf on a slide in a drop of water. Place the epidermal skin onto a clean, dry glass slide and add a few drops of glycerine. Cover the epidermal skin on Leaf Cross Section Under the Microscope. The second cut produces a thin slice or cross-section of leaf.
This takes practice.

Leaf cross section slide
|Leaf Structure Under the Microscope Preparation, Requirements and Observations Introduction Like any other multicellular living thing, leaf structure is made up of layers of cells. What do we expect to see inside the leaf?
With the sharp knife or razor blade and adult supervisioncut the leaf straight down the center. Monera is a biological kingdom that is made up of prokaryotes particularly bacteria. Here, the mesophyll section of the leaf contains two different type of cells including the palisade mesophyll elongated cells and the spongy mesophyll spherical or ovoid.
This includes the upper and lower epidermal cells flattened cells with the mesophyll layer in between. Because as grasses get blown around in the wind, they do not know which way is up!
